Do either of the HP reduction upgrades affect the hp of mobs in the challenges?  The base one affects non treasure monsters so I would think so...

Other one affects treausre spawns so would not seem like it would.  

But I dont know if either actually does, anyone know for sure?

The HP Reduction skill affects non-treasure monsters...which includes bosses in challenges.

The other skill only affects treasure monsters.
